Learning outcomes

The approach to security in the microbiology lab (containment levels with related facilities and activities). Bio-booths.
Principles and techniques of sterilization by dry heat and moist, sterilization by filtration, radiation sterilization, gas .

Microscopic examination of bacteria: preparation of slides, staining for bacteria, fungi, protozoa, direct observation or fresh. Cultivation of bacteria: culture media: basic, enriched, selective, differential, enrichment, transport, for susceptibility testing, for fungi.

Syllabus

The program approach to security in the microbiology lab (containment levels with related facilities and activities. The biosecurity cabins).
Principles and techniques of sterilization:
sterilization by heat dry and wet sterilization, by filtration sterilization, by radiation sterilization, by gas sterilization.
Techniques observation prepared by light microscopy:
optical microscope observation with lens dry, observation with immersion objective, observation brightfield, observation darkfield, observation in phase contrast .
Electron microscopy Principles
Microscopic examination of bacteria:
direct observation or fresh stains for bacteria, fungi, protozoa Cultivation of bacteria: Culture media: basal media, enriched, selective, differential, enrichment, transport, to antibiograms ,for fungi.

Culture media: basal media, enriched, selective, differential, enrichment, transport, to antibiograms for fungi.
Techniques and principles of sowing and isolation and identification of microorganisms:
the microbial isolation methods: general rules for isolation of the plate, the general isolation techniques, transfer of pure cultures, technical maintenance of pure cultures. Seeding techniques of biological samples.
Bacteria identification principles: identification based on the task biochemical identification based on serological and immunological techniques Methods of microbial counting methods for determining the sensitivity to antimicrobial agents: dilution and diffusion method
Techniques for the visualization of the antigen-antibody reaction:
enzyme immunoassay, immunofluorescence, latex agglutination test Notes on cultivation techniques in virology: cell cultures assessment of the growth of the virus in cell culture titration of virusess
Outline of molecular biology techniques applied to microbiological diagnostics:
Wester blot, Southern blot, PCR, qPCR.
Hints on quality control in microbiology.
